No. Once a monitor is created, you can put any number of assertions on it. We charge per monitor, per region, per check frequency.
A monitor across 3 regions counts as 3 monitors against your tier limit. The Pro 50-monitor cap, in practice, is ~16 monitors checked from 3 regions each.
At any time. Your monitors above the new tier limit are paused, not deleted. You can re-enable them by upgrading or removing some.
